Abstract. Tea pulp is traditionally used by ancient Indonesians as a treatment for the surface of the face, body or hair. In this study, antioxidant activity was tested using the DPPH free radical scavenging method (2,2-diphenyl-1-picrylhydrazyl), and the determination of total polyphenol levels using the Follin-Ciocalteu method. This study aimed to compare the antioxidant activities of ethanol extracts from black tea and green tea pulp and determination the levels of polyphenol compounds in black tea and green tea pulp. The samples used were ethanol extracts of black tea and green tea pulp, where the simplicia of tea pulp was obtained from the results of brewing based on SNI standards. Then the simplicia of tea pulp was extracted using maceration method using 96% ethanol. The results showed that the highest antioxidant activity of each sample was in the ethanol extract of black tea pulp with IC50 value of 1611.92 µg/mL and ethanol extract of green tea pulp was 6094.29 µg/mL. The highest results of the determination of total polyphenol levels were shown by the sample of green tea pulp ethanol extract of 46 % which was equivalent to 460 mg GAE/g sample, whereas black tea pulp ethanol extract sample was18.15 % which was equivalent to 181.5 mg GAE / g sample.Keywords: Tea pulp (Camellia sinensis (L.) Ampas teh secara tradisional sering digunakan masyarakat Indonesia jaman dahulu sebagai perawatan baik pada permukaan kulit wajah, tubuh, atau rambut. Pada penelitian ini dilakukan pengujian aktivitas antioksidan menggunakan metode peredaman radikal bebas DPPH (2,2-difenil-1-pikrilhidrazil), dan penetapan kadar polifenol total menggunakan metode Follin-Ciocalteu. Penelitian ini bertujuan untuk membandingkan aktivitas antioksidan ekstrak etanol dari ampas teh hitam dan teh hijau, serta menetapkan kandungan senyawa polifenol dalam ampas teh hitam dan teh hijau. Sampel yang digunakan adalah ekstrak etanol ampas teh hitam dan teh hijau, dimana simplisia ampas teh diperoleh dari hasil penyeduhan berdasarkan standar SNI. Kemudian simplisia ampas teh diektraksi dengan metode maserasi menggunakan pelarut etanol 96 %. Hasil penelitian menunjukan bahwa aktivitas antioksidan tertinggi dari tiap sempel terdapat pada ekstrak etanol ampas teh hitam dengan nilai IC50 1611,92 µg/mL dan 6094,29 µg/mL untuk ekstrak etanol ampas teh hijau. Hasil penetapan kadar polifenol total tertinggi ditunjukan oleh sampel ekstrak etanol ampas teh hijau sebesar 46 % yang setara dengan 460 mg GAE/g sampel, sedangkan untuk sampel ekstrak etanol ampas teh hitam sebesar 18,15 % yang setara dengan 181,5 mg GAE/g sampel.Kata Kunci: Ampas teh (Camellia sinensis (L.)
